چکیده

A multiple heart-cutting (mLC-LC) two-dimensional HPLC-UV achiral–chiral method for the direct analysis of branched-chain amino acids (BCAAs) in food supplements under environmentally friendly conditions was developed to cope with very well-known limited chemoselectivity chromatographic media enantioselective analysis. Both achiral and chiral methods were compliance main principles green chromatography. The performed isocratically an optimized ion-pair reversed-phase (IP-RP) based on a water/EtOH (95:5, v/v) mobile phase containing heptafluorobutyric acid (7 mM) as IP agent. characterized by appreciable performance validated before real sample. High recovery values all compounds (from 97% 101%) found interday evaluation. Additionally, low RSD% long-term period measured, range between 1.1% 4.8%. Still, LOQ value 0.06 mg/mL established compounds. quantitative commercial supplement revealed that BCAAs present amounts close those declared producer. carried out through application ligand-exchange chromatography (CLEC) approach, using O-benzyl-(S)-serine ((S)-OBS, 0.5 selector Cu(II) nitrate (0.25 metal source eluent. Resolution separation factor up 2.31 1.43, respectively, obtained. two systems connected six-port switching valve, mLC-LC confirmed absence D-enantiomers supplement, reported manufacturer’s label.

Catabolic Defect of Branched-Chain Amino Acids Promotes Heart Failure.

BACKGROUND Although metabolic reprogramming is critical in the pathogenesis of heart failure, studies to date have focused principally on fatty acid and glucose metabolism. Contribution of amino acid metabolic regulation in the disease remains understudied. METHODS AND RESULTS Transcriptomic and metabolomic analyses were performed in mouse failing heart induced by pressure overload. Suppressi...

Branched-Chain Amino Acids: Metabolism, Physiological Function, and Application Branched-Chain Amino Acid Enriched Supplements as Therapy for Liver Disease

Altered amino acid metabolism is a hallmark of liver disease, characterized by low levels of circulating BCAAs and elevated levels of circulating aromatic amino acids, and methionine. Although overwhelming evidence indicates that the incidence of complications of liver disease increases with malnutrition, the reported impact of nutritional therapy, specifically BCAA supplementation, on outcomes...

Branched-chain Amino Acids in Two Mentally Retarded Sibs

There is increasing interest in the importance of pyridoxine in brain disorders of childhood. Convulsions have occurred in infants receiving a pyridoxine (vitamin B6) deficient diet (Coursin, 1955), and Bessey, Adam, and Hansen (1957) have shown that some infants have a greater requirement than others for this vitamin. In the deficiency state, small amounts of vitamin B6 stop the convulsions, b...

Branched-chain amino acids suppress the cumulative recurrence of hepatocellular carcinoma under conditions of insulin-resistance

Branched-chain amino acids (BCAAs) reportedly inhibit the incidence of hepatocellular carcinoma (HCC) in patients with liver cirrhosis and obesity that is frequently associated with insulin resistance (IR). We previously reported that BCAAs exert a chemopreventive effect against HCC under IR conditions in rats. The aim of the present study was to examine the effect of BCAAs on the cumulative re...

Enzymic determination of branched-chain amino acids.

Transamination of branched-chain amino acids to (alpha-oxoglutarate, catalyzed by leucine aminotransferase coupled to the glutamate dehydrogenase reaction, is used in an enzymic assay for determination of branched-chain amino acids in serum and tissue homogenates. The coefficients of variation of the method within-day and day-to-day are 2.4 and 6.5%, respectively. Analytical recovery of physiol...
